No, I think they mean like a "comedy killer." That is, a joke, or routine that is so awkward, or otherwise unwelcome, that it kills any chance of making the crowd laugh for the rest of the night. Like when Rosanne Barr sang her farting rendition of the Star Spangled Banner before a ballgame, or when that jerk from the office tells a joke about "retards" after Bill from accounting just got done telling everyone how well his autistic ten-year-old is doing on the t-ball team.
